Standard iPSC Culture Conditions at the Facility
- mTeSR™1
- hESC-qualified Matrigel®
- clump passaging using enzyme-free EDTA-based ReLeSR™
Adaptation to Alternative Culture Conditions
The facility adapts hiPSC lines from diverse other culture conditions to standard conditions or vice versa. These include:
- KOSR (KnockOut™ serum replacement) based medium with FGF2 on inactivated mouse embryonal fibroblasts (MEFs) as feeder layer
- single-cell passaging using TrypLE™ Express
- mTeSR™ Plus
- StemFlex™ or Essential 8™ Flex
- Cellartis® DEF-CS™ culture system
- Laminin-521
- truncated Vitronectin (VTN-N)
The facility generates master and working cell banks of iPSC lines produced by the facility or from customer lines.
- Cryopreservation in Bambanker (unit: 10 vials)
- Storage in liquid nitrogen
- Post-thaw viability testing
